I have heard this book throuhg the recommendation from the twitter. So .. I searched for this book in the library. Fortuneateky, the book is available at Stang library, Faculty of Science, MU. I end up with borrowing this book to learn more about how to design a good experiment with reprducibility, even though, I have expereienced in doing research quite many years, still, I am curious why people recommend this book a lot through the twitter!

I have made a note, which will remind me what to do the research as well as tech my research fellows sometimes. The note will be updated time to time, depending on my reading speed.

Genome editing technology short note

เทคโนโลยีการตัดแต่งจีโนม (Genome editing technology) เป็นเทคโนโลยีที่อาศัยการกระตุ้นของการเกิด DNA double strand break แล้วเหนี่ยวนำให้เกิดกระบวนการซ่อมแซม DNA อันจะมีอยู่ 2 วีธีหลัก คือ  1. Homologous recombination (HR) ซึ่งต้องการ template ในการที่จะซ่อมแซมความเสียหายที่เกิดขึ้น ผลจากการซ่อมแซมดังกล่าวจะทำให้ได้สายของสารพันธุกรรมกลับมาเป็นอย่างเดิม (error-free) 2. Non-homologous end-joining (NHEJ) เป็นการซ่อมแซมแบบต่อสายของ DNA ที่ขาดเข้าโดยตรง ซึ่งกระบวนการต่อนั้นโดยปกติแล้วจะมีการเพิ่ม หรือการเติมของสารพันธุกรรมกลับเข้าไปในบริเวณที่เกิด DNA double strand break นอกเหนือไปจากนี้แล้ว อาจจะทำให้เกิดการ translocation ของตัว genome อันเนื่องมาจากการต่อกันที่ผิดของตัวแท่งโครโมโซม ดังนั้นแล้ววิธีการซ่อมแซมดังกล่าวส่งผลให้เกิดการเปลี่ยนแปลงของ genome (error-prone) ณ ตอนนี้เทคโนโลยีที่มีการกล่าวถึงมากที่สุด คือ Zinc finger, TALENs และ CRISPR/Cas ซึ่งถือว่าเป็นเทคโนโลยีสมัยใหม่ในการตัดแต่งจีโนม เทคโนโลยีทั้งสามดังกล่าวจะเกี่ยวข้องกับการทำให้เกิด DNA double strand break ก่อนโดยอาศัยกา
